The Dutch-British pair won 6-4, 6-4 against Spain's Marcel Granollers and Argentina's Horacio Zaballos.
It was the first Grand Slam victory for both. Koolhof had already won a major in mixed, Skupski had already won two there.
Granollers and Zeballos eliminated the German duo Kevin Krawietz/Tim Pütz in the semifinals.
As well as the trophy, Koolhof/Skupski take home a combined £600,000.
